چکیده

In vitro germplasms of ipecac (Cephaelis ipecacuanha Rich.), an important medicinal plant, maintained through reduced growth conditions for more than 12-years, were used as source material for micropropagation. MS with different combinations of Kn (2 mg/l), BAP (2 mg/l), 2iP (2-3 mg/l), NAA (0.2 mg/l) and adenine (40-100 mg/l) as additive was used to induce fresh multiplication of shoots from the nodal meristems and direct shoot bud initiation on the internodal segments. Complete plant regeneration has been achieved from such long term cultures. Regenerated plants maintained their phenotypic and chromosomal stability. Eighty per cent hardened plants, survived in the field condition, are growing well and 25% of them produced flowers within one year. Long term preservation through reduced growth conditions and successful regeneration of morphologically stable plants with stable chromosome numbers (2n = 22) from such long term cultures of ipecae plants.

Effect of L-dopa and L- Methionine Supplementation on Bioproduction of Ementine in Callus Cultures of Cephaelis Ipecacuanha

Callus cultures were induced from leaf segments of Cephaelis ipecacuanha on modified gamborg medium containing 2,4 - dichlorophenoxy acetic acid (2mg/1) Indole acetic acid (1mg/1), kinetin (1mg/1) and sucrose (20g/1). The biogenetic profile for alkaloids was established. The established callus cultures were transferred to modified gamborg medium supplemented with L- DOPA (40 mg/1) and L methion...

In vitro somatic embryogenesis from callus cultures of Cephaelis ipecacuanha

Callus was obtained from leaf explants of Cephaelis ipecacuanha on Murashige and Skoog (MS) basal medium supplemented with 0.5 mg/l Kin, 4.0 mg/l 2,4-D and 3% (w/v) sucrose [Murashige, T., Skoog, F., 1962. Reproductive studies on ipecac (Cephaelis ipecacuanha (Brot.) A. Rich; Rubiaceae): meiotic behavior and pollen viability.

Reproductive studies were carried out on Brazilian accessions of ipecac, Cephaelis ipecacuanha. Meiotic behavior was studied using the squashing technique. Irregular chromosome segregation in meiosis I and II, many sets of chromosomes in telophase II, micronuclei, incorrect cytoplasm division, incomplete cytokinesis and anomalous post-meiotic products, mainly polyads, were observed. The mean me...

Clonal diversity and conservation genetics of the medicinal plant Carapichea ipecacuanha (Rubiaceae)

The roots of the understorey shrub Carapichea ipecacuanha (ipecac) have medicinal properties, and the uprooting of wild plants has supplied most of the world demand for this species. Although under severe population decline, C. ipecacuanha lacks legal protection. In the wild, the aerial stems of ipecac clump together to form clusters with well-defined borders. Cluster size may range from severa...

Molecular cloning of an O-methyltransferase from adventitious roots of Carapichea ipecacuanha.

Carapichea ipecacuanha produces various emetine-type alkaloids, known as ipecac alkaloids, which have long been used as expectorants, emetics, and amebicides. In this study, we isolated an O-methyltransferase cDNA from this medicinal plant. The encoded protein (CiOMT1) showed 98% sequence identity to IpeOMT2, which catalyzes the 7'-O-methylation of 7'-O-demethylcephaeline to form cephaeline at ...
